沙棘果油及沙棘籽油對(duì)大鼠放射性皮膚損傷的影響

【摘要】  　　目的 觀察沙棘果油及沙棘籽油對(duì)大鼠放射性皮膚損傷的影響。方法 實(shí)驗(yàn)用深部X線治療機(jī)，每次以3000cGy的劑量，照射兩次，造成大白鼠臀部放射性皮膚損傷。待動(dòng)物癥狀明顯后，將Ⅱ度皮膚損傷的動(dòng)物隨機(jī)分為6組，即陰性對(duì)照組、沙棘果油高劑量組、沙棘果油低劑量組、沙棘籽油高劑量組、沙棘籽油低劑量組以及陽(yáng)性對(duì)照組。沙棘果油及籽油高劑量組，每天在損傷部位涂藥兩次，而其低劑量組每天涂藥一次，陰性對(duì)照組每天涂滅菌的注射用水一次，陽(yáng)性對(duì)照組每天涂Solcoseryl�睯elly軟膏一次。結(jié)果 肉眼觀察結(jié)果顯示，連續(xù)用藥10d后，沙棘果油、籽油及Solcoseryl�睯elly組動(dòng)物的局部紅斑、水腫明顯減輕，創(chuàng)面分泌物明顯減少，用藥2周后，上述癥狀明顯消退，與陰性對(duì)照組比較，明顯好轉(zhuǎn)；另外，沙棘果油、籽油及Solcoseryl�睯elly組的創(chuàng)面愈合時(shí)間明顯縮短。光學(xué)顯微鏡觀察結(jié)果顯示，沙棘果油、籽油及Solcoseryl�睯elly組的皮膚損傷明顯減輕，炎癥反應(yīng)亦明顯減輕，創(chuàng)面修復(fù)明顯。電子顯微鏡觀察結(jié)果顯示，陰性對(duì)照組的超微結(jié)構(gòu)明顯受損，而沙棘果油、籽油及Solcoseryl�睯elly組的損傷明顯減輕。結(jié)論 沙棘果油及沙棘籽油對(duì)大鼠放射性皮膚損傷有明顯的治療作用。

　　ABSTRACT: Objective  To observe the effects of sea buckthorn pulp oil and sea buckthorn seed oil on radiation skin lesions of rats. Methods  The experiment used deep x�瞨ay therapeutic machine to irradiate the rats twice, and the irradiation dosage was 3000cGy each time. This caused radiation skin lesions to rats�� buttock. After the symptoms became obvious, the rats suffering from Grade Ⅱ skin lesions were randomly divided into six groups: negative control group, high�瞕ose sea buckthorn pulp oil group, low�瞕ose sea buckthorn pulp oil group, high�瞕ose sea buckthorn seed oil group, low�瞕ose sea buckthorn seed oil group, and positive control group. The oil was applied to the wounded area twice a day for high�瞕ose pulp oil group and high�瞕ose seed oil group, and it was applied once a day for the low�瞕ose groups. For the negative control group, sterilized injection water was applied once a day, and Solcoseryl�睯elly ointment was applied once a day for the positive control group. Results  The results from naked eye observation showed that after medication for 10 consecutive days, the local erythema and edema of the groups using sea buckthorn pulp oil and seed oil and Solcoseryl�睯elly ointment abated, and the secretions of the wounded surface reduced considerably. After medication for two weeks, obvious disappearance of the above symptoms was observed; compared with the negative control group, the condition improved considerably. In addition, the healing　time of the wounded area of the rats was markedly shortened for the pulp oil groups, seed oil groups, and the Solcoseryl�睯elly group.Optical microscopic observation showed that the injuries in the pulp oil group,seed oil group,and the Solcoseryl�睯elly group were significantly reduced, inflammatory reaction was obviously reduced, and the restoration of the wounded areas was obvious. Electron microscopic observation showed that the ultrastructure of the negative group was obviously damaged, but that of the pulp oil groups, seed oil groups, and Solcoseryl�睯elly group reduced significantly. Conclusion  The above results suggest that sea buckthorn pulp oil and sea buckthorn seed oil have a marked therapeutic effect on radiation skin lesions in rats.

　　沙棘籽油及沙棘果油是從胡頹子科植物沙棘的籽及果肉中提取。據(jù)報(bào)道，沙棘油可用于治療潰瘍病、燒、燙傷等多種疾病[1]。張文祿等[2]報(bào)道沙棘油對(duì)腫瘤患者放射治療所引起的急性放射性皮炎、黏膜炎癥及非放射性的皮膚炎癥、潰瘍、傷口愈合不良等都有較好的療效。高力英等[3]報(bào)道沙棘油對(duì)患者放射性皮膚損傷有防治作用。本實(shí)驗(yàn)觀察了用超臨界萃取法提取的沙棘籽油及沙棘果油對(duì)大鼠放射性皮膚損傷的影響。

　　健康的SD大鼠，雌性，體重(200±20)g，由西安交通大學(xué)醫(yī)學(xué)院實(shí)驗(yàn)動(dòng)物中心提供。

　　1.2  藥品

　　沙棘籽油(sea buckthorn seed oil, SBSO)及沙棘果油(sea buckthorn pulp oil, SBPO)，用超臨界萃取法提取，由芬蘭土爾庫(kù)大學(xué)提供。Solcoseryl�睯elly軟膏，由瑞士素高大藥廠生產(chǎn)。
